Test ID: IH Granzyme B
Granzyme B
Immunohistochemistry Stain
Useful For

Identification of the neoplastic counterparts from human cytotoxic T lymphocytes (CTL) and natural killer (NK) cells.

Method name and description
  • Monoclonal Mouse Anti-Human Granzyme B, Clone GrB-7
  • Immunoperoxidase stain on formalin-fixed, paraffin-embedded (FFPE) tissue section
Reporting name

Granzyme B

Clinical information

Granzyme B, a 29 kDa monomer protein, is a member of a subfamily of serine proteases with chymotrypsin-like specificity. For CTL and NK cells to excert their cytolytic activity, the presence of cytoplasmic secretory granules containing granzymes and perforin are pivotal. After granule exocytosis, granzyme B is believed to enter the target cell through a perforin-derived transmembrane channel to induce DNA fragmentation and apoptosis. A high percentage of activated CTL’s, identified by anti-granzyme B, clone GrB-7, and present in tumour biopsies of patients with nodular sclerosis or mixed cellularity Hodgkin’s disease, has been reported to be a strong indicator for an unfavorable outcome.

Specimen type / Specimen volume / Specimen container
  • Specimen type: Well-fixed tissue in 10% neutral buffered formalin.
  • Submit 2 unstained (3-4 µm thick) paraffin embedded tissue section mounted on a clean positively charged glass slide  (sections to be cut within 6 weeks.) or formalin-fixed, paraffin-embedded (FFPE) tissue block.
Storage and transport instructions
  • Slides or blocks are stored in cork box at room temperature away from sun light and any source of heat.
  • Follow your local regulation shipping guidelines.
Specimen Rejection Criteria
  • Broken slides
  • Unlabeled slides with patient/case identification
  • Contaminated slides
  • Slides/paraffin blocks mismatch
  • Uncharged slides
