Test ID: Tissue culture
Tissue culture
Microbiological culture of tissues including biopsies and bone samples by conventional culture technique
Useful For

Infections of wounds ,tissues and biopsy specimens including bone.

Method name and description

Microscopy: Gram Stain
Culture: Aerobic and Anaerobic conventional culture technique

Reporting name

Tissue culture

Clinical information

A biopsy may be defined as a portion of tissue removed from the body for further examination. Tissue obtained whilst operating is particularly precious as the sampling procedure may not be repeatable. Ideally these specimens should be discussed with the laboratory prior to sampling to ensure that transport and processing are timely, and appropriate tests are performed.

Note: Tissue received as whole part for example: whole finger, entire toe, big chunk of tissue is not acceptable

Specimen volume

  • The specimen should ideally, be large enough to carry out all microscopy preparations and cultures.
  • Minimum specimen size will depend on the number of investigations requested. Numbers and frequency of specimen collection are dependent on clinical condition of patient.

Specimen container

  • Sterile leak proof container
Collection instructions / Special Precautions / Timing of collection

Collection instructions

  • The skin surface and surgical area are disinfected before collection, and the specimen is obtained aseptically by an invasive technique.
  • For small samples, add a few drops of sterile saline to keep moist.
  • Do not allow tissue to dry out.

Note: The portion of the biopsy specimen submitted for culture should be separated from the portion submitted for histopathology

Special Precautions

Do not place sample in formalin as it is unsuitable for microbiological investigations

Timing of collection

  • Prior to antimicrobial therapy where possible
Relevant clinical information to be provided
  • Antimicrobial therapy
  • Clinical condition
  • Site of infection

Storage and transport instructions
  • <2 hrs. at room temperature. Do NOT store
Specimen Rejection Criteria
  • Specimen sent in formalin
  • Tissue received as whole part for example: whole finger, entire toe, big chunk of tissue
  • Unlabeled/Mislabeled specimens
  • Improper container
  • Duplicate specimen within 72 hours
  • Quantity not sufficient (only culture performed- no microscopy done)
  • No relevant clinical details provided with the request. (Please note that where applicable, the relevant clinical details should reflect those on the patient’s electronic record at the time the order was placed).
Biological reference intervals and clinical decision values

Negative Microscopy report

No organisms seen

Positive Microscopy report

Organisms seen reported based on Gram stain result and morphology

Note: All STAT Gram stains called to Physician or Nurse in-charge, if positive.

Negative Culture report

  • No growth
  • Normal skin flora
  • Normal skin flora including coliforms
  • Mixed growth of coliforms

Positive Culture report

  • Significant organism identified and reported with antimicrobial susceptibilities
Factors affecting test performance and result interpretation
  • Improper collection
  • Delayed transport

Turnaround time / Days and times test performed / Specimen retention time

Turnaround time

Microscopy:

  • STAT specimens: 1 hour
  • Routine specimens:24 hrs

Note:

  • No Gram stain done for rectal and intestinal biopsies e.g. colonic biopsy, duodenal aspirates or bone.

Culture: 2-5 days

Note: Preliminary results are released as early as the presumptive identification is available

Note:

  • Duodenal aspirate or biopsy - No culture, for ova and parasite exam only.
  • Oesophageal swab / biopsy - for fungal culture only.

 Days and times test performed

  • 24/7

Specimen retention time

  • 7 days after final report
