Test ID: DIF IgM
Direct Immunofluorescence IgM
Direct Immunofluoresence Stain
Useful For

Demonstration of human IgM in tissues and assessment of pattern, extent and intensity of glomerular staining. Assess presence and pattern of IgM deposits in other sites (Tubular basement membrane, blood vessels, interstitium).

Method name and description
  • Polyclonal Rabbit Anti-Human IgM/FITC.
  • Immunofluorescence stain on frozen tissue section.
Reporting name

IgM Direct Immunofluorescence

Clinical information

IgM FITC reacts with the m-chain of human IgM. Traces of contaminating antibodies have been removed by solid-phase absorption. It is essential to supplement clinical findings and histopathology in the diagnosis of autoimmune diseases. It permits early diagnosis, treatment, and subsequent monitoring of disease activity in patients with these potentially life-threatening disorders.

Aliases

ImmunoglobulinM/FITC

Specimen type / Specimen volume / Specimen container
  • Specimen type: Submit 2 unstained frozen sections (2 µm thick) mounted on a clean positively charged glass slide fixed in acetone to be sent dry , or fresh tissue placed in normal saline.
  • One slide will be stained for IgM antibody and the other slide will be stained for the negative control (the use of the negative control is to check for non-specific signal and false positive results).
Storage and transport instructions
  • Slides should be packaged well to avoid damage if they are dropped or crushed.
  • You can ship your specimens in suitable slide box placed in dry ice.
  • Follow your local regulation shipping guidelines.
Specimen Rejection Criteria
  • Broken slides
  • Unlabeled slides with patient/case identification
  • Contaminated slides
  • Slides mismatch
  • Uncharged slides
  • Improper fixative and storage
Factors affecting test performance and result interpretation
  • Fixation time (FT)
  • Fixative Type
  • Storage temperature
  • Age of the cut sections
  • Section thickness
Turnaround time / Days and times test performed / Specimen retention time
  • Turnaround time for the test in platform: 1 hour
  • Days and times test performed: Daily (Sunday to Thursday)
  • Time: 0700H-1400H
  • Shelf-life of the frozen section slides: ≤2 weeks
